According to the Associated Press, the fully loaded 136-car Union Pacific coal train derailed east of Jefferson City, Missouri. It had originated in Wyoming and was traveling to Kentucky.
Local newspaper the News Tribune said the derailment occurred about 2.30pm local time Saturday and while initial reports said 40 cars had been overturned, a final count Monday morning came to 50 cars.
The eastern and western lines were reportedly reopened early Monday.
Work to clean and repair the derailment area will take several more days due to the remote location of the incident as well as a 16-inch snowfall obstacle.
The cause of the derailment is yet to be determined, the AP said.
